Genetic and Molecular Targets in Lymphoma
BCL-6, CCND1, c-myc and BCL-2 are cell-cycle regulatory proteins. These proteins can be generated by chromosomal translocations like t(8:14), t(14:18), t(11:14) and t(3:14), and impact prognosis individually. Occasionally, concomitant expression of either two (double hit) or three (triple hit) of these proteins is observed. This event is conventionally assumed to confer a more aggressive disease course with poor outcomes. Protein overexpression apart from chromosomal breaks can also be due to gene amplification or mutations in promoter regions coding for genes, which when analyzed by molecular testing has aided in prognostication.
